Screening of Biotechnologically Useful Enzymes in Halophiles/Haloalkaliphiles from Coastal Areas of Gujarat: Molecular and Functional Characterization for Industrial Applications. Major 2008 In this collaborative project between our lab, IIT Delhi, and Rajkot University in Gujarat, the halophilic bacteria isolated from salt lakes and coastal areas of Gujarat were characterized for their biochemical properties at IIT, and the relevant genes were cloned in our lab. Identification and Functional Analysis of Molecular Interactions Involved in the Development of Reproductive Floral Organs in Rice Major 2009 The candidate genes identified in the first phase were characterized for their function during stages of reproductive development. Specifically, genes involved in male gametophyte development and seed development were targeted. This work led to the publication of six research papers and the completion of two Ph.D. degrees. Characterization of Molecular Components Involved in Pollen Abortion During High Temperature Stress Major 2015 This study identified a set of genes putatively involved in conferring differential resilience to heat stress in the rice varieties N22 (tolerant) and IR64 (susceptible). The investigation employed a comparative transcriptomic analysis using a microarray-based assay to examine gene expression profiles under controlled heat stress conditions. Analysis of the transcriptional responses revealed that the stress avoidance pathway is likely activated in the heat-tolerant variety N22. Notably, this response involved an accelerated progression of pollen development, enabling the completion of reproductive processes prior to the onset of severe stress, which may underlie the enhanced thermotolerance observed in N22 in contrast to the susceptibility of IR64. Characterization of Gene Regulatory Networks and Transcriptional Mechanisms that Control Male Gametophyte, Functional Characterization of Genetic and Epigenetic Regulatory Networks Involved in the Reproductive Development in Rice Major 2015 In this project, we successfully delineated the functions of several genes implicated in male gametophyte development. The encoded proteins were found to be crucial for the timely degradation of the tapetum and for pollen exine formation. Additionally, tissue-specific promoters with activity restricted to the tapetum and/or developing pollen were identified and functionally characterized. These promoters were subsequently employed to generate male sterile lines in rice. Molecular-physiological Characterization of Epigenetic Components Affecting Plant Development Under Drought and High-Temperature Stress Major 2015 This project primarily facilitated travel grants for the Indian and Egyptian teams, enabling mutual visits to each other's institutions to share expertise and foster long-term collaborative relationships. As part of the exchange, members of our laboratory visited Cairo once, while our Egyptian counterparts visited the University of Delhi on two occasions. These visits enabled fruitful exchanges of ideas and technical know-how, particularly in the areas of plant transformation, stress physiology, and the characterization of stress-responsive elements. Development of a Cell-line-based Starch Bio-factory System by Manipulating Components of Starch Regulatory and Biosynthetic Machinery in Tobacco BY-2 Cells Major 2020 This project successfully established a starch bio-factory system in tobacco BY-2 cells by co-expressing a rice transcription factor OsMADS29 along with three tobacco genes involved in starch biosynthesis. This genetic modification led to a substantial increase in starch accumulation—from 1.5% in wild-type cells to approximately 25% in transgenic cell lines. The results demonstrate effective metabolic reprogramming and highlight the utility of OsMADS29 as a cross-species regulator. The developed platform offers a promising, scalable approach for high-yield starch production in plant cell cultures, with potential applications in industrial biotechnology and sustainable bioresource generation. Deciphering the Role of Translational Regulation in Embryo and Endosperm Differentiation During Early Seed Development in Rice Major 2023 In this ongoing project, we have successfully completed RNA-seq, miRNA-seq, and polysomal RNA-seq analyses of rice developing seeds at 1, 4, and 7 days after pollination (DAP). These datasets provide a comprehensive temporal landscape of transcriptional and translational changes during early seed development. The next phase involves proteomics analysis, which, together with existing data, will be integrated through a multi-omics pipeline. This will enable the identification of specific mRNA elements that mediate translational regulation of key developmental genes. Ongoing since 19-10-2023 (1 years, 7 months, 25 days)
